摘 要:本研究探讨索拉非尼(sorafenib,SOR)联合柔红霉素(daunorubicin,DNR)对K562细胞株的增殖活性及凋亡的影响。采用MTT法检测K562细胞株在索拉非尼单药及联合柔红霉素作用下增殖的抑制效应,根据两药相互作用指数(coefficient of drug interaction,CDI)评价两药相互作用性质,并用Hoechst33258荧光染色及流式细胞术检测细胞凋亡情况。结果表明:索拉非尼单药及联合柔红霉素对K562细胞株有显著的抑制效应,而且两药联合应用具有协同作用,联合组凋亡细胞增加。结论:柔红霉素和索拉非尼联合作用于白血病细胞株K562呈现明显的协同抑制作用。The aim of this study was to investigate the effect of sorafenib combined with daunorubicin on leukemic k562 cell line. The inhibitory effect of sorafenib alone and its combination with daunorubicin on K562 cell proliferatin was detected by MTF method; the synergistic effect was measured by CDI (coefficient of drug interaction ); the apoptosis of K562 cells was observed by flow cytometry with Hoechst 33258 staining. The results showed that the sorafenib alone or its combination with daunorubicin could significantly inhibit K562 cell prohferation and the combination of both drugs displayed synergistic effect on K562 cells, meantwhile the apoptotic cells increased. It is concluded that the combination of sorafenib and daunorubicin has a obviously synergestic inhibitory effect on leukemic cell line K562.
