摘  要:目的探讨缺血/再灌注大鼠脑和血浆中α-突触核蛋白(α-Synuclein,α-Syn)含量的变化。方法将30只雄性Wistar大鼠用随机数字表法分成假手术对照组(10只)和缺血/再灌注组(20只)。用线栓法制作大脑中动脉梗阻(middle cerebral artery occlusion,MCAO)模型,缺血1.5h后,再恢复灌注24h;用Western blotting法和ELISA法检测大脑缺血半暗带、梗死核心区和血浆中α-Syn的含量变化。结果缺血半暗带α-Syn表达量显著增加,而梗死核心区和血浆中α-Syn无显著性变化。结论缺血/再灌注可增加缺血半暗带中α-Syn的含量。Objective To explore the change of α-synuclein(α-Syn) in rat brain and plasma after ischemia/reperfusion. Methods Thirty adult male Wistar rats were randomly divided into 2 groups:sham operation group(10 cases) and ischemia/reperfusion group(20 cases). A model of middle cerebral artery occlusion(MCAO) was induced by the intraluminal suture for the latter group. After 1.5 h ischemia,reperfused for 24 h,then the level of α-Syn in the plasma and penumbra,infarct core and their controls were measured by Western blotting and ELISA method. Results The level of α-Syn was increased significantly in the penumbra,but did not change obviously in the infarct core or plasma. Conclusion α-Syn was increased in the penumbra of brain after ischemia/reperfusion.
