摘 要:目的:研究咬合重建对大鼠三叉神经节降钙素基因相关肽(calcitonin gene-related peptide,CGRP)及CGRPmRNA表达的影响。方法:Wistar雄性大鼠48只,随机均分为3个实验组及正常对照组。实验组动物间断磨除右上、下颌磨牙牙冠至龈下,有2组分别第3周、第9周停止磨牙,任其自行萌出,恢复咬合关系。双侧三叉神经节(trigeminal ganglia,TG)切片行CGRP免疫组织化学反应(SABC法)和原位杂交反应。光镜观察拍片,并用I mage Pro Plus5.1图像分析软件进行测定。SPSS10.0软件行统计分析。结果:单侧咀嚼实验组咀嚼侧和非咀嚼侧TG内CGRP免疫阳性神经元百分比与对照组比较显著降低(P<0.01),其非咀嚼侧明显低于咀嚼侧(P<0.01),而CGRPmRNA阳性神经元百分比显著增高(P<0.01),非咀嚼侧明显高于咀嚼侧(P<0.01)。晚期恢复咬合实验组TG内CGRP和CGRPmRNA表达情况与单侧咀嚼实验组一致。结论:早期恢复咬合关系TG内CGRP和CGRPmRNA表达可恢复正常,晚期恢复咬合关系其表达不能恢复正常,CGRP和CGRPmRNA参与了单侧咀嚼引起的颞颌关节病的病理变化过程。Objective:To study the expression of calcitonin gene-related peptide(CGRP)and CGRPmRNA in trigeminal ganglia(TG)in rats with occlusal reconstruction,and to discuss the mechanism of temporomandibular disorders.Methods:48 Wistar male rats were randomly divided into 3 experimental groups and 3 control groups,with 8 rats in each group.The right molars of maxillary and mandibular of rats in experimental groups were ground to the gingival margin level without occlusal contact.The occlusal contact was recovered by stoping grounding molars of the rats.The section of trigeminal ganglia underwent the immunohistological and situ hybridization study.Light microscope and microscopic photo analysis software were employed to detect the percentage of CGRP and CGRPmRNA positive neurons in frozen section of TGs in 48 rats.SPSS10.0 statistical software was used for statistical analysis.Results:The percentage of CGRP positive neurons in TG of unilateral chewing experimental group was significantly decreased compared with control group(P0.01).The decreased extent on non-chewing side was much higher than that on the chewing side(P0.01).The percentage of CGRPmRNA positive neurons was higher on both chewing and non-chewing sides of the unilateral chewing experimental group than that in control group(P0.01),and that on non-chewing side was much higher than that on the chewing side(P0.01).The result of late-stage occlusal reconstruction experiment group was as same as that of unilateral chewing experimental group.Conclusion:The expressions of CGRP and CGRPmRNA in TG can recover after the early occlusal reconstruction and not after the late-stage occlusal reconstruction.CGRP and CGRPmRNA might participate in the histopathologic mechanism of temporomandibular disorders.
