摘 要:目的观察缺血性脑损伤后大鼠缺血皮质区microRNA 210的表达变化规律,探讨mir-210对脑缺血后血管新生的可能影响。方法实验分为缺血后1d、3d、7d组和假手术组(n=5),缺血组采用线栓法制作大脑中动脉阻塞模型,于缺血后1、3、7d处死大鼠,取缺血皮质区用于实验;假手术组仅暴露动脉。提取各实验组脑缺血皮质区的miRNA,Real time PCR比较各实验组mir-210的表达水平。结果脑缺血后mir-210表达显著上调,缺血后1d、3d、7d分别为假手术组的7.2±0.56、20.1±0.87和20.3±0.76倍(P<0.05)。结论脑缺血后mir-210表达显著上调,mir-210可能在促进脑缺血后血管新生过程中起重要作用。Objective To investigate the potential roles of mir-210 in angiogenesis after brain stroke by de-tecting its expression changes in impaired cortex after cerebral ischemia in rats.Methods The experiment rats was divided into four groups: 1,3,7 days post-ischemia groups and sham group.Middle cerebral artery occlusion(MCAO) rat models were made by obstructing MCA using a single strand nylon thread.Rats were killed at 1,3,7 days after ischemia,and the impaired cortex were extracted for examination.The sham group was not operated as control.MiRNAs were isolated from ischemic cortex and real time PCR was used to determine the expression of mir-210.Results Mir-210 expression was significantly increased in ischemic cortex after ischemia compared with sham group,with 7.2±0.56(1d),20.1±0.87(3d) and 20.3±0.76(7d) fold changes respectively.Conclusions The expression of mir-210 shows significantly increased in ischemic cortex after ischemia,indicating a potential role in enhancing angiogenesis after brain ischemia.
