摘  要:为研究气态甲醛对外周血白细胞DNA的损伤作用,选用SPF级昆明种纯系雄性小鼠作为研究对象,用浓度为0.5 mg/m2、1.0 mg/m2、3.0 mg/m3气态甲醛对小鼠进行连续动态染毒72 h,染毒结束后,观察外周血白细胞的DNA断裂和DNA-蛋白质交联的状况.实验表明:气态甲醛能诱导小鼠外周血白细胞的DNA断裂,在各染毒浓度组与对照组均有及显著差异,在0.5 mg/m3浓度组DNA断裂效应比1.0 mg/m2,3.0 mg/m3浓度组高;1.0 mg/m2,3.0 mg/m3浓度组出现明显的DNA-蛋白质交联作用.研究结果表明:气态甲醛暴露对小鼠外周血白细胞的DNA有明显的损伤作用.For studying on DNA damage of peripheral blood leukocytes by gaseous formaldehyde, SPF-class Kun-Ming male mice were used to be exposed to gaseous formaldehyde in different concentrations: 0. 5 mg/m^-3,1.0 mg/m^-3 and 3.0 mg/m^-3. After 72 hours continuous dynamic inhalation, comet assay and adding protease K methods were used to observe DNA breakage and DNA-protein cross-linking situation of peripheral blood leukocytes. The results showed that gaseous formaldehyde induced DNA breakage in peripheral blood leukocytes, particularly at low concentrations of the apparent DNA- protein cross-linking role. And it proved that gaseous formaldehyde has significant damage effect on DNA in peripheral leukocytes of mice.
