摘 要:目的研究丹红注射液对氯化锂-匹罗卡品点燃慢性颞叶癫痫(temporal lobe epilepsy,TLE)大鼠海马神经元及GFAP表达的干预作用。方法建立氯化锂-匹罗卡品点燃颞叶癫痫大鼠模型,将50只造模成功的颞叶癫痫大鼠随机分为颞叶癫痫组(生理盐水10ml/kg腹腔注射)及丹红治疗组(丹红注射液10ml/kg腹腔注射),另选10只大鼠作为正常对照组;每组大鼠随机分为5个不同时间点:24h、3d、7d、15d及30d。采用尼氏染色及免疫组化染色,检测每组大鼠不同时间点海马CA1区神经元存活数目及GFAP表达。结果与正常对照组相比,颞叶癫痫组大鼠海马CA1区存活神经元数目明显减少,海马GFAP表达显著增多(P<0.05);与颞叶癫痫组相比,丹红治疗组大鼠海马CA1区存活神经元数目明显增多,海马GFAP表达显著减少(P<0.05)。结论丹红注射液可以减轻颞叶癫痫大鼠海马神经元脱失及减少GFAP表达,有助于减轻大鼠发作程度及阻断慢性颞叶癫痫的形成和发展。Objective To investigate the effects of Danhong injection on the neuronal loss and glial fibrillary acidic protein(GFAP)expression of hippocampus CA1 in the temporal lobe epilepsy(TLE)rats induced by lithium-pilocarpine.Methods The TLE rats models were established with lithium chloride and pilocarpine intraperitoneally.50 TLE rats were randomly divided into 2 groups:TLE group(NS 10 ml/kg,intraperitoneal injection),and Danhong injection intervention group(Dan hong injection 10 ml/kg,intraperitoneal injection).In addition,10 rats were assigned to the normal control group.Each group were randomly divided into five different time points:24 h,3 d,7 d,15 d and 30 d.Nissel staining was conducted for valuating the survival neurons in hippocampus CA1 in each groups.The changes of GFAP were examined by immunohistochemistry staining.Results Compared with normal control group,rats in TLE group showed fewer survival neurons(P〈0.05)and more expression of GFAP in hippocampus CA1(P〈0.05);Compared with TLE group,Danhong injection intervention group had significantly improved the quantity of survival neurons(P〈0.05)and showed fewer expression of GFAP in hippocampus CA1(P〈0.05).Conclusion Danhong injection is helpful to relieving the extent and blocking the formation and development of seizures in rats with chronic TLE by alleviating the neuronal loss and reducing the expression of GFAP in hippocampus CA1.
