皮肤瘢痕组织中PTEN、Caspase-3、Survivin基因表达的临床意义

摘　　要：目的:探讨皮肤瘢痕组织中PTEN、Caspase-3、Survivin基因表达的临床意义。方法:用原位杂交方法对35例皮肤瘢痕组织、15例正常皮肤组织的石蜡包埋标本进行PTENmRNA、Caspase-3mRNA、SurvivinmRNA检测,并对数据进行统计学处理。结果:SurvivinmRNA在皮肤瘢痕组织中的表达明显高于正常皮肤组织(p<0.01),PTENmRNA、Caspase-3mRNA在皮肤瘢痕组织中表达明显低于正常皮肤组织(p<0.01)。PTENmRNA与SurvivinmRNA共阳性(6/35)17.14%,一致性为(13/35)37.14%,二者的表达呈负相关;PTENmRNA与Caspase-3mRNA共阳性(12/35)34.29%,一致性为(28/35)80.00%,二者的表达呈正相关。结论:PTENmRNA、Caspase-3mRNA、SurvivinmRNA基因在皮肤瘢痕组织中的异常表达、凋亡与增殖的失衡可能是皮肤瘢痕形成的重要原因。Objective：To study the gene expression of PTEN, Caspase -3,and Survivin in skin scar tissues and its clinical significance. Methods：Paraffin -embedded specimens of 35 cases of scar tissue and 15 cases of normal skin tissues were detected by PTENmRNA,Caspase - 3mRNA,SurvivinmRNA by in situ hybridization,and statistical analysis was performed by SPSS system. Results：The result showed that the expression of SurvivinmRNA in skin scar tissues was higher than those in normal skin tissues （p 〈0.01 ） ,while PTENmRNA and Caspase - 3mRNA in scar tissues were lower （ p 〈 0.01 ）. The positive expression rates of PTENmRNA and SurvivinmRNA were 17. 14% ,and the concordance rates were 37.14%. The expression of PTENmRNA and SurvivinmRNA showed inverse relation. The positive expression rates of PTENmRNA and Caspase- 3mRNA were 34.29%, and the concordance rates were 80.00%. Positive correlations between PTENmRNA and Caspase -3 mRNA were noted. Condusion： The abnormal expressions of PTENmRNA, Caspase - 3 mRNA,and SurvivinmRNA in scarand the imbalance of apoptosis and proliferatian may be the important factors of skin scar tissues.
