摘 要:目的探讨α-硫辛酸(LA)对高糖和H2O2引起的ECV304细胞氧化损伤的保护作用。方法用不同剂量的α-硫辛酸0.25,0.5,1mmol/L预处理ECV30424h,再用30mmo/l葡萄糖作用24h,流式细胞术检测细胞线粒体ROS含量和线粒体膜电位。用上述三组剂量的LA预处理ECV304 24h,再用100μmol/LH2O2作用2h,单细胞凝胶电泳检测细胞DNA氧化损伤情况。结果经不同浓度LA预处理后,ECV304的ROS含量与对照组比显著下降(P<0.01),且随剂量增加有下降趋势;线粒体膜电位值显著地升高(P<0.05或者P<0.01),且随剂量增加有升高趋势。对于H2O2造成的细胞DNA断裂损伤,在加入不同浓度LA后,细胞拖尾率、尾长和Olive尾矩等指标均有显著性下降(P<0.05或者P<0.01)。结论一定浓度范围内的α-硫辛酸对高糖和H2O2引起的ECV304细胞的氧化损伤具有保护作用。Objective To investigate the protective effects of α-lipoic acid (LA) against oxidative damage induced by hyperglycemia and hydrogen peroxide in ECV304 cells.Method ECV304 cells were pretreated with different concentra-tions of LA 0.25,0.5,1 mmol/L for 24h.The cells were treated with 30mmol/L glucose for 24h to induce oxidative damage and with 5.6mmol/L glucose as control.Reactive oxygen species (ROS) and mitochondrial membrane potential were detected by flow cytometer.The cells were also treated with 100μmol/L H2O2 for 2h after pretreated with LA and the non-pretreated with LA as positive control.DNA damage was determined by single cell gel electrophoresis (SCGE).Results The concentrations of ROS in LA pretreated groups were significantly decreased than that of 30mmol/L glucose treated group.And the mitochondrial membrane potentials in LA pretreated groups were significantly increased than that of 30mmol/L glucose treated group.The indices of DNA damage cells,tail DNA,tail length,Olive tail moment (OTM) were all significantly decreased in LA pretreated group than those of H2O2 treated group.Conclusion α-Lipoic acid might protect ECV304 cell against oxidative damage induced by hyperglycemia and hydrogen peroxide.
