伴有淋巴结转移性声门上型喉癌的蛋白质组学研究

摘　　要：目的比较分析有无淋巴结转移性声门上型喉癌组织的差异表达蛋白质。方法应用固相pH梯度胶双向电泳技术分离不同转移潜能的声门上型喉癌组织总蛋白质,行银染色,Image master 2D软件分析双向凝胶电泳图谱,确定其差异表达蛋白质点。结果获得25种差异表达蛋白质点,可能与声门上型喉癌淋巴结转移有关;与无淋巴结转移性喉癌比较,有淋巴结转移的喉癌组织中20种蛋白质表达上调,5种蛋白质表达下调。结论有无淋巴结转移的声门上型喉癌组织蛋白质表达谱存在明显差异,这些差异表达蛋白质可能与声门上型喉癌的淋巴结转移特性有关。Objective To comparatively analyze the differentially expressed proteins in the tissues of supraglottic carcinoma among two groups of cases either with or without neck lymph node metastasis based on a study on clinical samples. Methods Two dimensional electrophoresis （2-DE） was used to separate the differentially expressed protein spots from the total protein extracted from the tissues of supraglottic carcinoma, either with or without neck lymph node metastasis, based on the techniques of solid phase gradient pH gel electrophoresis, silver staining and computer-assisted image analysis system by the soft ware Image master 2D. Results Twenty five spots of differentially expressed protein in total were separated from the total protein of supraglottic carcinoma tissues, possibly associated with the metastatic potentiality to neck lymph node. When compared with that of supraglottic carcinoma without neck lymph node metastasis, 20 kinds of differentially expressed protein were up-regulated and 5 were down-regulated markedly in the supraglottic carcinoma with neck lymph node metastasis. Conclusions There are significant differences in the expressive spectrum of proteins in the tissues of supraglottie carcinoma, either with or without neck lymph node metastasis. These differentially expressed proteins may be closely associated with the potentiality of neck lymph node metastasis for supraglottic squamous cell carcinoma.
