非小细胞肺癌组织中PDGF-A mRNA表达的意义及与淋巴管计数的关系

摘　　要：目的研究非小细胞肺癌(non-small cell lung cancer,NSCLC)组织中血小板源性生长因子A(platelet-derived growth factor-A,PDGF-A)的mRNA表达和淋巴管计数,探讨其临床病理意义及相互关系。方法 70例NSCLC手术切除标本行常规石蜡包埋切片,原位杂交法检测PDGF-A mRNA表达,SP免疫组化法检测D2-40表达,并进行淋巴管计数。结果 70例NSCLC组织中PDGF-A mRNA阳性49例,阳性率为70%。TNM分期Ⅰ期、无淋巴结转移、组织学高中分化的病例,PDGF-A mRNA阳性率及D2-40标记淋巴管计数明显低于Ⅱ/Ⅲ期、有淋巴结转移、组织学分化低的病例(P<0.01),而鳞癌与腺癌比较无统计学差异(P>0.05)。PDGF-A mRNA阳性病例中的淋巴管计数明显高于阴性病例的淋巴管计数(P=0.000)。结论 PDGF-A mRNA表达和淋巴管计数与NSCLC的发生、进展及转移等生物学行为有密切关系,PDGF-A可能具有促NSCLC组织淋巴管生成的作用。Objective To study the expression of platelet-derived growth factor-A（PDGF-A） mRNA and the relationship of the lymph vessel count,and to investigate their clinic-pathological significances in non-small cell lung cancer（NSCLC）.Methods In situ hybridization and SP immunohistochemistry were performed to detect the expression of PDGF-A mRNA and lymph vessel count respectively in the routinely paraffin-embedded sections of specimens from 70 patients with NSCLC.Results The positive rate of PDGF-A mRNA in NSCLC were 70%（49/70）.The positive rate of PDGF-A mRNA and the lymph vessel count were significantly lower in cases of TNM stage Ⅰ,without lymph node metastasis and histological high/moderate differentiation than those in UICC TNM stage Ⅱ/Ⅲ,with metastasis and histological low differentiation（P〈0.01）.There was no significant difference on PDGF-A mRNA expression between the squamous carcinoma and adenocarcinoma（P〉0.05）.The counts of lymph vessels were significant higher in PDGF-A positive cases than those of the negative expression in NSCLC（P〈0.001）.Conclusions The expression of PDGF-A mRNA and the lymph vessel count might be important markers in reflecting the progress,biological behavior and metastatic status of NSCLC.PDGF might promote the lymphangiogenesis in NSCLC.
