摘 要:目的探讨氯化锂(LiCl)对大鼠急性心肌缺血-再灌注(I-R)损伤的保护作用。方法 60只SD大鼠随机平均分为4组:假手术(A)组,I-R(B)组,LiCl干预(C)组,L-精氨酸甲酯(L-NAME)+LiCl干预(D)组。实验前7d开始,D组腹腔注射L-NAME30mg·kg-1.d-1。经左侧肋间切口进胸。C和D组术前经颈静脉置管注射LiCl15mg/kg,B、C、D组行冠状动脉左前降支(LAD)结扎术,缺血60min,再灌注240min。记录心率和心电图变化,实验结束时检测血浆肌酸激酶同工酶(CK-MB)、羟丁酸脱氢酶(HBDH)、丙二醛(MDA)和超氧化物歧化酶(SOD)、一氧化氮合酶(NOS)活力及心肌组织中MDA含量、SOD活力;在光镜和电镜下比较组织形态学变化。结果与B组相比,C组SOD活力显著增高,MDA含量、再灌注心律失常发生及血HBDHCK-MB水平显著降低(P<0.05);光镜和电镜下C组心肌病理损害较B组明显减轻。结论 LiCl可能通过减轻氧化应激途径对大鼠心肌急性I-R损伤起保护作用。Objective To investigate the protective effects of lithium chloride(LiCl)on myocardial ischemia-reperfusion(I-R)injury in rats.Methods Sixty SD rats were randomly divided into four groups of sham operation(group A),I-R(group B),I-R+LiCl(group C),and I-R+L-NAME+LiCl(group D)with 15 rats each.L-NAME 30mg·kg-1·d-1 was administered intraperitoneally in group D since 7 days before experiment.LiCl 15mg/kg was given in groups of B and C before ischemia via jugular vein.I-R was induced by ligating left anterior descending artery for 30 minutes and then reperfusing for 240 minutes in groups of B,C and D.Heart rate and ECG were recorded.Plasma levels of α-hydroxybutyrate dehydrogenase(HBDH),creatine kinase isoenzyme-MB(CK-MB),activity of nitricoxide synthase(NOS),myocardial and plasma content of malondialdehyde(MDA)and activity of superoxide dismutase(SOD)were detected at the end of experiment.Histological observation of myocardium was performed under light microscopic and transmission electron microscopy.Results Compared with group B,the activity of SOD increased and MDA content in myocardium and plasma reduced(P0.05).Plasma levels of HBDH and CK-MB and incidence of reperfusion-induced arrhythmias were significantly dropped in group C as well(P0.05).Pathologic evaluation showed that myocardial lesion was less in group C than that in group B.Conclusion LiCl may protect I-R-induced myocardial injury through attenuating myocardial oxidative stress.
