摘 要:目的动态观察慢性盐负荷对自发性高血压大鼠(SHR)胸主动脉胶原含量及TGFβ1表达的影响,并探讨其可能的机制。方法 80只SHR随机分为2组:4%(质量分数)高盐饮食组(高盐组,n=45)和0.4%(质量分数)正常盐饮食组(对照组,n=35),持续干预16周。每隔4周监测血压、尿量及尿钠。分别于实验第0、4、8、12、16周时随机分批处死SHR(每次每组6只),取胸主动脉行Masson染色,观察病理变化并计算主动脉胶原容积分数(CVF),免疫组化法测定主动脉管壁转化生长因子β1(TGFβ1)的蛋白表达。结果①实验第4、8、12、16周,高盐组SHR主动脉CVF均较对照组增高[(8.02±1.21)%vs.(6.87±2.15)%,(10.35±2.07)%vs.(8.21±1.02)%,(16.20±1.58)%vs.(15.31±2.13)%,(19.37±1.816)%vs.(18.7±1.9)%,P<0.05]。②实验第4、8、12周时高盐组SHR主动脉TGFβ1的表达均较对照组增高[(8.02±1.21)%vs.(6.87±2.15)%,(10.35±2.07)%vs.(8.21±1.02)%,(16.20±1.58)%vs.(15.31±2.13)%,P<0.05]。结论慢性盐负荷可使SHR胸主动脉的胶原含量增加,而局部TGFβ1含量的升高可能是其致胶原含量增加的重要原因之一。Objective To make a dynamic observation on the effect of chronic salt loading on collagen content in thoracic aorta and the expression of TGFβ1 in spontaneously hypertensive rats (SHR) and investigate the possible mechanism. Methods SHRs (80 in number,male,4-5 weeks old) were assigned randomly to receive 4% (g/kg) high salt (HS groups,n=45) and 0.4% low salt (LS groups,n=35) for 16 weeks. Blood pressure,urine volume and urinary sodium excretion were measured every four weeks. These rats were sacrificed randomly at the start of this study,4th,8th,12th and 16th week,6 rats in each group each time. The aorta was collected and dyed with Masson to observe the pathological change of thoracic aorta under microscope; microholography system was used to calculate collagen volumetric fraction (CVF) of the aorta. Expression of TGFβ1 in SHR thoracic aorta was measured with immunohistochemical method. Results ① CVF in thoracic aorta was increased significantly in HS group compared with that in LS group at 4th,8th,12th and 16th week (P0.05). ② Compared with that in LS group,the expression of TGFβ1 in HS group was increased at 4th,8th and 12th week (P0.05). Conclusion Chronic salt loading can increase collagen content in thoracic aorta of SHR. Local TGFβ1 increase induced by salt loading in SHR is probably involved in the development of collagen increase.
