肿瘤坏死因子对人滋养层干扰素α分泌的影响

摘　　要：目的和方法：本文采用人妊娠６～８周胎盘组织块的体外培养方法，通过检测干扰素免疫活性和抗病毒活性研究肿瘤坏死因子α（ＴＮＦα）对人滋养层干扰素（ＩＦＮ）分泌的影响。结果：ＴＮＦα可显著促进人滋养层ＩＦＮα的分泌，并呈剂量和时间依赖关系。同时各浓度的ＴＮＦα还可促进βＨＣＧ的分泌，并呈一定的剂量依赖关系。此外，我们还证明ＩＮＦα可显著抑制人滋养层细胞的生长，而ＴＮＦα则无明显作用。结论：ＴＮＦα可通过促进人滋养层干素α和βＨＣＧ的分泌。Aim and Methods:To investigate the effects of TNFα on the secretion of human trophoblast IFN in the first trimester placental explants in vitro.Results: TNF α signifcantly enhanced the production of IFN α in both dose and time dependent manners,as showed by the immunoactivity and antiviral activity of IFN. On the other hand, TNF α significantly stimulated the secretion of β HCG in a dosedependent manner. Furthermore, IFN α but not TNF α showed a suppression on the growth of primary human placental trophoblasts.Conclusion: TNF α may affect the function of human placenta by activating human trophoblast IFN α production.
