鼠三型肝炎病毒诱导不同小鼠肝炎模型肝脏自然杀伤细胞基因差异表达谱研究

摘　　要：目的:研究重型肝炎和轻、中度慢性肝炎肝脏NK(自然杀伤)细胞基因表达谱的差异,探讨重型肝炎和慢性肝炎疾病中NK细胞的生物学功能。方法:采用MHV-3(鼠三型肝炎病毒)诱导的Babl/cJ小鼠暴发型肝炎模型和C3H/Hej小鼠感染慢性化模型,利用Affymetrix小鼠全基因组芯片检测两种小鼠模型肝脏NK细胞的基因表达谱差异。结果:在小鼠45038条全基因组探针中,有3708条表现上调,有统计学意义的有736条,同时有4509条基因表现下调,有统计学意义的有866条。较之感染慢性化模型,在暴发型肝炎中上调的基因主要集中在物质代谢过程、细胞活动调节过程、细胞相互作用过程以及免疫学过程。较之暴发型肝炎模型,感染慢性化模型上调的基因主要集中在特异性免疫过程、细胞凋亡过程、癌症发生过程以及纤维组织增生过程等。结论:NK细胞参与MHV-3诱导的暴发型肝炎和慢性肝炎的发生发展过程,但与之相对应的,NK细胞也参与持续感染过程的慢性化的疾病过程中,其内在机制值得进一步探讨。Objective：To elucidate differential gene expression profiles in the background of whole mice genome between severe hepatitis and mild/moderate chronic hepatitis,and discuss the function of hepatic NK cells in severe hepatitis.Methods：Two mice hepatitis models,Balb/cJ fulminant hepatitis model and C3H/Hej chronic hepatitis model,which were induced by MHV-3 were adopted in the research.The hepatic NK cells were separated by MACS and reserved for the Affymetrix mouse genome 430 2.0 array.Results：Among the whole 45038 genomic probes,there were 3708 upregulated probes in which 736 probes had distinguished difference,while there were 4509 downregulated probes in which 866 probes had remarkable difference.Comparing with the chronic hepatitis model,the function of upregulated genes in fulminant hepatitis model focused on the process of material metabolism,regulation of cellular activities,interaction between cells and immune response.Meanwhile,the function of upregulated genes in chronic hepatitis model concentrated in the process of specific immune response,apoptosis,cancer development and fibroplasia compared to that in the fulminant hepatitis model.Conclusion：NK cells participated in the pathogenesis of MHV-3 induced fulminant hepatitis,however,NK cells also participated in the persistant infection process in chronic hepatitis,the inside mechanism of which was deserved furtherly discussed.
