Fhit、Survivin基因在宫颈病变中的表达与高危型HPV感染的相关性研究

摘　　要：目的探讨Fhit、Survivin在宫颈正常组织、宫颈上皮内瘤变和宫颈癌中的表达与HR-HPV感染的相关性,并探讨其在宫颈癌发生发展中的意义。方法应用SABC法检测Fhit和Survivin基因的表达,并于术前采用HC-Ⅱ法检测HR-HPV感染情况。结果 Fhit和Survivin表达成负相关(P<0.05)。Fhit表达与宫颈癌患者的年龄、临床分期、肿瘤大小、病理分级和淋巴转移均无相关性(P>0.05)。Survivin表达与宫颈癌病理分级、临床分期及淋巴转移相关(P<0.05)。Survivin在宫颈癌中的表达和HR-HPV感染率均高于正常宫颈组织(P<0.05),而Fhit在宫颈癌中的表达与以上两者成负相关(P<0.05)。结论 Fhit表达缺失发生在CIN期,Survivin则参与了宫颈癌发生、侵袭和转移的过程,并与HR-HPV感染导致的宫颈上皮细胞异常增生有关。Objective To study the expression of Fhit, Survivin in normal cervical tissue, cervical intraepithelial neoplasia （CIN） and cervical cancer. To explores their correlation with HR - HPV infection and significance in cervical cancer＇s occurrence and development. Methods The expressions of Fhit and Survivin were detected by immunohistochemistry SABC assay ,and HR - HPV infection were detected by HC - Ⅱ before operation. Results Fhit expression was negatively associated with Survivin expression （ P 〈 0. 05 ）. The expression of Fhit show no correlation with patients age, clinical staging, tumor＇s size, patho - grade and lymph node metastasis in cervical cancer （P 〉 0.05 ）. The expression of survivin had significant with clinical staging, patho - grade and lymph node metastasis in cervical cancer （ P 〈 0. 05 ）. The expression of Survivin in cervical cancer and HR - HPV infection rate were higher than the normal cervical tissue, but the expression of Fhit was negatively related with both of them （ P 〈 0. 05 ）. Conclusion Expression loss of Fhit happened in CIN. Survivin takes part in cervical cancer＇s tumorigenesis, invasion and metastasis, and related with the abnormal proliferation of cervical epithelium tissue by HR - HPV infection. The abnormal expression of Fhit and Survivin play a synergetic role in the occurrence and development of cervical cancer.
