摘 要:目的喉癌是喉部最常见的恶性肿瘤,喉癌的发病率目前有逐年提高的趋势。近年来的研究表明,奥曲肽对许多实体肿瘤及正常组织有抗增生作用,还具有抑制肿瘤细胞增殖、诱导肿瘤细胞凋亡的作用。本实验旨在研究奥曲肽对喉癌Hep-2细胞增殖和凋亡的作用及其机制的初步探讨。方法通过MTT比色试验和测定细胞生长曲线研究奥曲肽对Hep-2细胞增殖的作用,经流式细胞仪检测研究奥曲肽对Hep-2细胞凋亡和细胞周期的影响。结果奥曲肽浓度在0.05μg/ml-20μg/ml范围内对Hep-2细胞均有抑制作用,48 h时最显著,但血清组与无血清组比较,抑制率明显下降(P<0.05)。无血清组奥曲肽浓度在0.1μg/ml的奥曲肽抑制作用最为显著,为24.67%(P<0.01,vs con-trol)。在奥曲肽作用下72 h内,Hep-2细胞的生长曲线有明显下降(P<0.05),奥曲肽浓度在0.05μg/ml和0.1μg/ml范围内,Hep-2细胞生长曲线较低平。经流式细胞仪检测,Hep-2细胞出现G0/G1阻滞,0.1μg/ml的奥曲肽最显著(73.97±0.48)%(P<0.05);实验组G2/M期细胞比例亦有下降(P<0.05 vs control);而实验组与对照组S期细胞的比例无显著性差异(P>0.05)。结论本研究结果提示奥曲肽抑制Hep-2细胞增殖可能是通过诱导G0/G1阻滞和诱导细胞凋亡而实现的,为喉癌的综合治疗提供了理论依据。Objective Laryngeal carcinoma is the most common malignant tumor occurring in larynx and the incidence of laryngeal cancinoma shows an increasing trend recent years. According to the recent researches, somatostatin analog octreotide can not only contribute to resisting hyperplasy aiming at many solid tumors and normal tissues, but also inhibiting proliferation and inducing apopto- sis of malignant cells. So our research is to study the effect of octreotide on cell proliferation and apoptosis in laryngeal carcinoma cell Hep-2 and its mechanism.Methods The proliferation of Hep-2 cells was determined by MTT assay and cell growth curve. Ceil cycle blockage and apoptosis were detected by flow cytometry. Results The proliferation of Hep-2 cells was inhibited significanfly by octreotide (0.05,0.1,0.5,1.0,10.0, and 20.0 μml), and the inhibition of 0.1 μg/ml octreotide was most significant, 24.67 % (P 〈 0.01, vs control). Comparing with the Hep-2 cells of serum-free group, the inhibition of serum group decreased significantly (P 〈 0.05) .And the growth curves of Hep-2 cell depressed significantly (P 〈 0.01, vs control); during 72 hours, the growth curves of Hep-2 cell became flatter with the concentration from 0.05 μ/ml to 0.1 μg/ml. The results of flow cytometry: Hep-2 cells showed G0/G1 blcokage and apoptosis was also induced, 0.1 μg/ml the most: (73.97 ± 0.48) % and (2.27 ±0.15) % ( P 〈 0.05, vs control) ,the cells of G2/M phase decreased ( P 〈 0.05 vs control) ;but the ceils of S phase did not have great change( P 〉 0.05 vs control). Conclusion Octreotide can directly inhibit Hep-2 cells proliferation by inducing ceil cycle blockage and apoptosis, so the knowledge of octreotide' s effect on ceil cycle will be helpful for the formulation of the treatment for laryngeal carcinoma.
