摘  要:目的:研究大鼠肺纤维化发生中白细胞介素-1β(IL-1β)及其I型受体的动态表达。方法:气管内注入博莱霉素制备大鼠肺纤维化模型,观察肺组织病理变化,测定肺组织中羟脯氨酸浓度,免疫组化法检测其中IL-1β及其Ⅰ型受体的含量。结果:病理切片显示模型组大鼠肺泡间隔明显增宽,较多纤维母细胞增生及炎性细胞浸润,后期肺组织实变;羟脯氨酸的含量随造模时间的推移逐渐升高,于28d时达峰值;模型组大鼠在第7天时肺组织中IL-1β及其Ⅰ型受体的表达积分达到峰值,14d时显著下降,但仍高于正常组(P<0.01),28d时基本回到正常水平。结论:IL-1β及其Ⅰ型受体在肺纤维化的早期显著升高,提示其可能在肺纤维化发生的早期起重要作用。Objective:To study the changes of Interleukin-1 beta(IL-1β) and its type Ⅰ receptor in pulmonary fibrosis in rats.Methods:The pulmonary fibrosis model was established by intratracheal injection of bleomycin with rats;investi gate the lung tissue pathological changes in the rats of pulmonary fibrosis,and assay the content of hydroxyproline in lung tissue,test the contents of IL-1β and its receptor and in the lung tissue using the method of immunohistochemistry.Results:Pathological evidence of the M-group rats demonstrated that the alveolar compartment companied with massive in flammatory cell invasion and a number of myofibroblast proliferation became more thick.The content of hydroxyproline was increasing as time goes,and achieved peak on the 28th day.IL-1β and IL-1RⅠin lung tissues in model-group shew a highest expression scoring on the 7rd,and reduced on the 14th day,and equal to the normal-group almost on the 28th day.Conclusion:IL-1β and the type I receptor for IL-1β in pulmonary fibrosis raise to the peak early and briefly,and play an important role in the earlier period of pulmonary fibrosis.
