摘  要:目的:研究胎盘生长因子(PIGF)在乳腺癌组织中的表达及其临床意义。方法:分别采用实时荧光定量PCR(real-time quantitative,Q-PCR)和免疫组织化学方法检测56例乳腺癌组织和72例癌旁组织中PIGF的表达。结果:Q-PCR检测乳腺癌组织中PIGFmRNA的表达水平为0.987±0.171,相应癌旁组织中PIGFmRNA的表达量为0.265±0.114;免疫组化检测乳腺癌组织中PIGF蛋白阳性率为80.4%(45/56),相应癌组织中PIGF蛋白阳性率为12.5%(8/72);癌组织中PIGF的表达均显著高于癌旁组织(P<0.05)。PIGF的高表达与淋巴结转移及Nottingham预后指数有相关性(P<0.05),但与乳腺癌的病理类型、临床分期及ER状态无相关性。结论:PIGF在乳腺癌组织中高表达,且与乳腺癌的发生、发展及临床预后密切相关。PIGF很可能在乳腺癌的发病机制中起着重要的作用。OBJECTIVE:To investigate the expression and clinical significance of placenta growth factor (PlGF) in human breast cancer.METHODS:The PlGF mRNA expression was examined quantitatively by using real-time quantitative polymerase chain reaction on a cohort of human breast cancer tissues (n=56) and corresponding adjacent tissues (n=72),and the immunohistochemical method was used to estimate the expression of PIGF protein.RESULTS:By using Q-PCR,the expression levels of PIGF mRNA in breast cancer and adjacent tissues were 0.987±0.171 and 0.265±0.114,respectively;by using immunohistochemistry,the expression rates of PIGF protein in breast cancer and adjacent tissues were 80.4%(45/56) and 12.5%(8/72),respectively.PlGF expression was dramatically increased in breast cancer tissues compared with the normal background breast tissue (P〈0.05).The overexpression of PIGF in breast cancer was correlated with tumour-node-metastasis and Nottingham prognostic index (P〈0.05),but not with other clinicopathological factors,including pathological type,clinical stage and ER.CONCLUSION:PlGF is over-expressed in breast cancer tissues and significantly correlates with clinical prognosis,and may have a pivotal role in breast cancer development and disease progression.
