摘 要:目的:检测多药耐药相关蛋白(MRP)基因在乳腺癌组织中的表达水平,并和谷胱甘肽S-转移酶-pi(GST-π)的表达作相关性比较,探讨其在临床的应用价值。方法:用定量RT-PCR技术,检测了55例乳腺癌组织中MRP和其中40例GST-πmRNA的表达水平,并和术前化疗比较分析。结果:MRP表达的阳性率为76.4%,高表达占18.2%,中低表达各占23.6%和34.5%。MRP和GST-π有40%的共表达率,虽然MRP表达阳性率及水平和术前化疗关系不明显,但化疗后二者表达有正相关趋势。结论:MRP在乳腺癌中普遍表达,以中低表达为主,其和GST-π的共表达可能和化疗药物诱导有关。Objective: The gene expression of multidrug resistance associated protein (MRP) in breast cancer and its relation with the glutathione S transferase π (GST π) were studied.Methods:The expression of MRP gene in 55 cases of breast cancer was measured by quantitative PCR, and the level of GST π was also determined in 40 cases.The correlation between chemotherapy and MRP expression was analysed as well. Results: Positive expression was shown in 76.4% of the cases. Of these, high level expression of MRP was found in 18.2% , median level in 23.6% and low level in 34.5%. Coexpression of MRP and GST π was seen in 40% of breast cancer. In addition,no relationship was found between the amount of MRP and effect of chemotherapy, but the level of MRP was higher than those who received chemotherapy before surgery.Conclusion:MRP is a major factor associated with drug resistance in breast cancer. MRP and GST might be induced by chemotherapeutic agents simultaneously.
