摘  要:目的建立针对在生化鉴定结果不准确的情况下,利用分子生物手段对病原菌进行鉴定的方法。方法将分离出的一株细菌经过普通的形态学观察和氧化酶试验,用肠杆菌科生化试纸条(API20E)做生化鉴定。并利用细菌通用引物对细菌的16S—23S rDNA间区进行PCR扩增,测序,利用BLAST在线同源性查询软件查询所测片段的属性。结果 API20E生化鉴定结果可能是河流弧菌,BLAST比对结果为嗜水气单胞菌(GenBank accession No:FJ013143)。结论传统的表型分类在细菌鉴定中已逐步与各种基因型分类水平的细菌鉴定方法相结合,在实际工作中应注意有效使用。[Objective]To establish the method of identification of pathogenic bacteria using molecular biology methods under the situation of that the result of biochemical identification is not accurate.[Methods]After the morphological observation and the oxidase test,the bacteria isolated was identified with API20E,then 16s-23S rDNA intergenic spacer(IGS) with the bacteria-specific primers was PCR-amplified and sequenced. The properties of fragment were checked by BLAST online software system for homology check.[Results]The result of API20E biochemical identification showed that the pathogenic bacteria might be Vibrio fluvialis. While the analysis result by BLAST indicated it was Aeromonas hydrophila(GenBank accession No:FJ013143).[Conclusion]The traditional phenotypic classification of bacteria has gradually combined with various genotype classification and identification of pathogens. It's an effective way to combine these 2 methods for practical work.
