摘 要:目的探讨采用荧光原住杂交(FISH)技术检测乳腺癌患者中的Her2基因与免疫组织化学(IHC)检测Her2基因的结果的一致性以及导致两者差异的可能原因;了解荆州地区乳腺癌患者中Her2基因的分布。方法以FISH方法,对55例免疫组织化学分别为(3+),(2+),(1+)和阴性(-)的乳腺癌新鲜石蜡切片标本进行Her2基因的检测。结果55例标本中,有16例采用FISH方法检测出Her2基因的表达;12例Her2基因表达(3+)的标本中11例检测出Her2基因;7例Her2基因表达(2+)的标本中5例检测出Her2基因;10例Her2基因表达(1+)的标本中未检测出Her2基因;26例Her2基因表达(-)的标本中未栓测出Her2基因。结论初步筛查Her2基因状态可以首选免疫组织化学方法,对于IHC(2+)的标本,应该行FISH确诊。Objective To detect the expression of Her2 gene by Fluorescene in situ hybridization (FISH) and Immunohistochemistry(ICH) ,and to evaluate the concordance and correlation between FISH and IHC. Methods FISH for Her2 gene expression status was performed using PathVysion DNA probe kit on the archival paraffin-embedded sections of breast cancer tissues from 55 patients with immunohlstochemical staining scores of(3+),(2+),(1+) and 0. Results 11 of the 12 patients with score(3+) by IHC were positive for Her2 by FISH. 5 of the7 patients with score(2+) by IHC showed Her2 gene amplification. All the patients with scores (1 +) and 0 by IHC failed to show amplification. Conclusion Immunohistochemistry is useful as an initial screening tool for Her2 expression status. Patients with score (2 +) by IHC should undergo FISH testing as well.
