摘  要:利用聚丙烯酰胺凝胶(Bio-Gel P-2)柱分离高纯度低聚果糖制备单一组分,结果表明,在1.6 cm×100 cm的玻璃柱上最佳的层析条件是洗脱速度0.2 mL/min、上样量0.5 mL(样液质量浓度0.4 g/mL)、柱温40℃。以Megazlaae公司的标准品为对照,经HPLC和MS分析,证明层析分离所得组分峰4、峰3和峰2分别是蔗果三糖、蔗果四糖和蔗果五糖。而且经HPLC面积归一化法定量分析,证明分离产品蔗果三糖、蔗果四糖、蔗果五糖蔗和果六糖的含量分别为99.91%、99.94%、99.33%和97.89%,表明其纯度很高,可以作为低聚果糖定量分析的标准品使用。The separation of high-purity fructooligosaccharide(FOS) monocomponents by polyacrylamide column was investigated.The results showed that optimized chromatographic conditions were as following: a flow rate of 0.2 mL/min,a sample volume of 0.5mL(o.4g/mL),a column of 1.6×100 cm at temperature 40 ℃.The products obtained from the separation of FOS monocomponent(peak 5,4 3) were analysed by HPLC and MS using Megazlaae's standards as reference.The results indicated that the products included 1-kestose,1,1-kestotetraose and 1,1,1-kestopentaose,respectively.The purity of 1-Kestose to 1,1,1,1-kestohexose measured by quantitative analysis by HPLC peak area normalization were 99.91%,99.94%,99.33% and 97.89%,respectively.This indicated that the purity was high enough to be used as standard for quantitative analysis of fructooligosaccharides.
