摘 要:目的研究肾皮质细胞膜转化生长因子β(TGFβ)受体(TβR)在大鼠糖尿病发病中的作用。方法观察单侧肾切除后建立的糖尿病大鼠4周后血糖、血胰岛素、血肌酐水平、体重、肾重、肾组织蛋白含量及血浆、肾皮质、髓质ACE活性的变化;利用RT-PCR检测肾皮质中TβRⅠ、ⅡmRNA表达,Western杂交检测肾皮质细胞膜TβRⅠ蛋白表达。结果同对照组相比,糖尿病大鼠尽管体重进行性下降,肾重和肾组织蛋白含量却明显增加,肾皮质中ACE活性也明显增加。RT-PCR检测肾皮质内TβRⅠ、ⅡmRNA表达分别增加1.12和4.0倍;Western杂交显示肾皮质细胞膜TβRⅠ蛋白表达增加1.10倍。结论糖尿病大鼠肾皮质中TGFβR表达明显上调,可能是高糖和局部RAS活性增加共同作用的结果。Purpose To study role of transforming growth factor β receptor(TβR)in the renal cortex from diabetic rats. Methods Blood glucose,serum insulin,serum creatinine,body weight,kidney weight and renal protein content as well as ACE activity in the plasma,renal cortex,medullawere observed after 4 weeks of SRZ injection.mRNA expression of TβRⅠ and Ⅱwere measured by quantitative reverse transcription polymerase chain reaction(RT-PCR);Protein expression of TβRⅠ was determined by western blot analysis in kidney cortex cell membrane. Results Diabetic rats developed hyperglycemia ( P <0.01),decreased serum insulin ( P <0.01),elevated serum creatinine ( P <0.05),body weight loss ( P <0.01),kidney hypertrophy ( P <0.05) and increased ACE activity ( P <0.05) in renal cortex.These changes were associated with 1.12 and 4.00 fold increase in TβRⅠ and Ⅱ mRNA expression,respectively.Protein expression of TβRⅠ was increased by 1.10 fold in renal cortex cell membrane. Conclusions Up-regulation of expression of TGFβR in renal cortex may be associated with hyperglycemia and locally active RAS.
