Anti-apoptotic and Neuroprotective Effects of Edaravone on Neurological Tissue after Acute Spinal Cord Injury in Rats

摘  要:目的探讨依达拉奉对急性脊髓损伤大鼠的抗细胞凋亡和神经保护作用机制。方法将120只SD大鼠随机分为3组:假手术组(n=20)、对照组(n=50)和治疗组(n=50),采用改良的Allen法,制成中度脊髓损伤大鼠模型。假手术组行椎板切除,不用任何药物;治疗组注射依达拉奉3 mg.kg-1,每隔12 h重复一次,直至相应时点;对照组仅给予等量生理盐水。各组于术后6、24、48、72、120 h 5个时点分别取样检测脊髓组织含水量及丙二醛(MDA)的含量;TUNEL法检测各组脊髓中的神经细胞凋亡数;免疫组化检测各组脊髓组织Bcl-2基因表达。结果治疗组各时点脊髓组织含水量、MDA含量及细胞凋亡数均低于对照组(P<0.05或P<0.01),Bcl-2蛋白表达显著高于对照组(P<0.01)。结论依达拉奉能降低脊髓组织MDA含量,减轻脊髓水肿,上调Bcl-2表达,抑制细胞凋亡,减轻脊髓继发损伤,从而达到其对大鼠脊髓损伤的抗细胞凋亡和神经保护作用。Objective To investigate the anti-apoptotic and neuroprotective effects of edaravone on neurological tissue after acute spinal cord injury in rats.Methods A total of 120 health male SD rats were randomly divided into false-operation group(n=20),ASCI control group(n=50) and ASCI treatment group(n=50).Rats of false-operation group were only done eaminetomy none used drugs;Rats of treatment group were injected edaravone 3 mg·kg-1 per 12 th to end point;Rats of control group were only used equivalent NS.according to 6、24、48、72 h and 120 h after model creation.The model of moderate spinal cord injury in rats was established by Allen method.Spinal tissue water content and MDA were measured.respectively according to 6、24、48、72 h and 120 h after model creation.Neuronal apoptosis and the expression of Bcl-2 were monitored by TUNEL and immunohistochemistry respectively.Results The spinal tissue water content,MDA and apoptosis were significantly decreased in ASCI treatment group as compared with ASCI control group(all P0.05 or P0.01),while the expression of Bcl-2 in ASCI treatment group was markedly higher than that in ASCI control group(P0.01).Conclusion Edaravone may reduce MDA and edema,and increase expression of Bcl-2 as well as inhibit apoptosis,thus decrease the secondary spinal cord injury and shows anti-apoptotic and neuroprotective effects on neurological tissue after acute spinal cord injury.
