摘  要:肌内脂肪含量(intramuscular fat,IMF)被认为是影响畜禽肉质风味的主要因素,而作为影响畜禽肌内脂肪沉积的候选基因,心脏型脂肪酸结合蛋白(heart-type fatty acid-binding proteins,H-FABP)基因对鸡肌内脂肪的沉积起着重要的作用。本实验选择H-FABP基因的3个变异(SNP或插入缺失)对华南农业大学杏花鸡×隐性白洛克鸡全同胞家系F2代资源群进行了标记-性状关联分析,并利用实时荧光定量PCR(quantita-tivereal-timePCR,qRT-PCR)技术对不同组织、不同日龄及不同基因型的H-FABP基因mRNA表达水平上的差异进行了研究。结果表明,G+926A位点多态性与胸肌粗脂肪含量呈极显著相关(P<0.01);14bp插入缺失位点与鸡肉质等性状关联分析的结果都是差异不显著(P>0.05),仅与70日龄和84日龄体重等生长性状的相关性达到极显著相关水平(P<0.01);C+2851T位点与腿肌粗脂肪含量显著相关(P<0.05)。qRT-PCR及单倍型分析结果及关联分析结果相一致,进一步表明H-FABP基因可以做为IMF等肉质性状的侯选基因。The heart fatty acid-binding protein gene (H-FABP) is regarded as one candidate gene that affected intramuscle fat (IMF) deposition, and generally thought to be a major factor that influences meat quality of livestock. Three variations(SNPor in/del) in thisgene were genotyped in a F2full sib ofXinhua×White Recessive Rock reference population, aiming at discovering their associations with chicken fatty traits. The expression patterns in different tissues, day-old, genotypes of H-FABP gene were further studied. The results showed that 1) the G+926A polymorphism wasverysignificantlyassociated with fat content ofbreast muscle (P0.01); 2) the 14 bp in/del was very significantly associated with body weight at the age of 70 days and 84 days (P0.01), while no associations of this polymorphism with fat traits were observed at significant level; 3) the C+2851T polymorphism was very significantly associated with fat content of leg muscle (P0.01). The results of qRT-PCR and haplotype were consistent with association study, this further suggested that H-FABP gene may be a candidate gene for chicken meat quality traits, such as IMF.
