摘 要:目的观察并检验尿苷三磷酸(UTP)对在体大鼠脑缺血再灌注损伤的保护作用。方法健康成年雄性SD大鼠100只,随机分为5组(n=20):假手术组、生理盐水组及G10、G30、G90组。除假手术组外,均采用线栓法制备大鼠中脑缺血再灌注损伤模型。G10、G30及G90组分别在大鼠大脑中动脉缺血30min后以微量输注泵通过尾静脉持续给予UTP10、30及90μg/kg,输注速度为5ml/(kg.min),假手术组及生理盐水组给予生理盐水注射,各组总药液容量均为1ml/kg。再灌注24h后评定各组大鼠神经功能缺损评分(NDS),然后从每组中随机抽取8只大鼠测定大脑半球含水量;从假手术组、生理盐水组及G90组中各取2只大鼠脑组织标本用于电镜观察组织超微结构;每组另取10只大鼠,通过TTC染色观察脑梗死体积。结果 UTP对大鼠脑缺血再灌注损伤具有保护作用,G90组的保护作用最明显,其NDS评分(0.7)明显低于生理盐水组(1.8,P<0.01),梗死侧大脑半球含水量(85%)明显高于生理盐水组(82.1%,P<0.01),梗死体积占整个脑组织的比例(16.9%)明显低于生理盐水组(30.5%,P<0.01)。电镜观察显示,与生理盐水组比较,G90组的细胞凋亡明显减少。结论 UTP对大鼠脑缺血再灌注损伤具有保护作用。Objective To explore the protective effect of uridine 5'-triphosphate (UTP) on cerebral ischemia reperfusion injury in rats.Methods One hundred adult male SD rats were randomly divided into five groups (20 each):sham operation group,saline control group,G 10 group,G 30 group and G 90 group.Ischemia was induced by intraluminal suture embolism of mesocephalic artery (MAO) of the rats in all the groups except sham operation group.UTP solution was delivered through an indwelling tail venous catheter via microinfusion pump 30min after the occlusion of mesocephalic artery at a rate of 5ml/(kg·min),and rats in G 10,G 30 and G 90 group received 10,30 and 90μg/kg UTP,respectively.Rats in sham group and saline control group received normal saline.The total fluid volume was the same,1ml/kg,in all groups.Neurological deficit score (NDS) was determined 24h after reperfusion.Eight SD rats were then randomly selected from each group,and water content of cerebral hemispheres was measured.Two rats from each of sham operation group,saline control group and G 90 group were selected for studying the ultrastructure of brain tissue by electron microscopy.Infarct volume was determined by 2,3,5-triphenyl-tetrazolium chloride (TTC) staining with another 10 rats in each group.Results The protective effect of UTP was determined on cerebral ischemia-reperfusion injury in rats,and in G 90 group showed the most significant protective effect.For the rats in saline group and G 90 group,NDS score was 0.7 and 1.8 (P〈0.01),water content of infarct cerebral hemisphere were 85% and 82.1% (P〈0.01),percentage of infarct volume vs total brain were 16.9% and 30.5% (P〈0.01),respectively.Electron microscopy showed less neural cell death in G 90 group than in saline control group.Conclusion UTP affords a certain protective effect on cerebral ischemia reperfusion injury in rats.
