摘　　要：目的研究胰岛素样生长因子Ⅰ型受体(IGF-IR)激酶抑制剂NVP-AEW-541及阿霉素联用对肝癌细胞株smmc-7721的治疗效果。方法 NVP-AEW-541与阿霉素单独和联合应用肝癌细胞株smmc-7721后,MTT法检测细胞生长抑制率,流式细胞术检测细胞凋亡,Western blot检测p27、抗凋亡蛋白Bcl-2及促凋亡蛋白Bax的表达。结果 NVP-AEW-541和阿霉素均可诱导smmc-7721细胞凋亡。与单独使用相比,两药联用对smmc-7721细胞抑制作用有明显提高(P<0.05),细胞凋亡率增加(P<0.05),对增加Bax、p27的表达及抑制Bcl-2的表达更明显。结论 NVP-AEW-541和阿霉素均可抑制smmc-7721细胞的增殖、诱导细胞凋亡,在一定浓度范围内,呈量-效关系。Objective To explore the proliferation-suppressing and apoptosis-inducing effects of NVP-AEW-541 combined with doxorubicin on smmc-7721 cells.Methods The smmc-7721 cells were treated with NVP-AEW-541（insulin-like growth factor recepter kinase inhibitor） and doxorubicin alone or in combination.The cell inhibition rate was evaluated by MTT assay,apotosis rate by flow cytometery,expressions of p27,Bax and Bcl-2 were detected by Western blot.Results Both of NVP-AEW-541 and doxorubicin induced the apoptosis of smmc-7721 cells（P〈0.05）with elevated expressions of Bax and p27 and decreased expression of Bcl-2.Conclusion Both of NVP-AEW-541 and doxorubicin,especially combined use,can suppress the proliferation and induce the apoptosis of smmc-7721 cells.
