摘 要:背景:溃疡性结肠炎(uc)的黏膜损伤与氧化应激相关。谷胱甘肽(GSH)是细胞内最重要的抗氧化剂,γ-谷氯酰半胱氨酸合成酶(γ-GCS)是其合成的关键酶。目的:探讨UC患者结肠黏膜中γ-GCS表达。方法:选取2007年8月~2009年12月北京军区总医院的30例UC患者,以26例健康者作为对照,应用免疫组化法检测结肠黏膜中γ-GCS的表达:结果:γ-GCS主要表达于腺体细胞胞质内,UC组γ-GCS表达显著低于对照组(53.3%对92.3%,P〈0.01)。结论:UC患者结肠黏膜中γ-GCS表达显著降低,说明其存在氧化应激失衡,且GSH合成减少可能与γ-GCS低表达有关.Background: Mucosal injury in ulcerative colitis (UC) is relevant with oxidative stress. Glutathione (GSH) is the most important antioxident and γ-glutamylcysteine synthetase (γ-GCS) is the key enzyme in the synthesis of GSH. Aims: To investigate the expression of γ-GCS in colonic mueosa of UC patients. Methods: Thirty patients with UC from August 2007 to December 2009 at The Militai'y General Hospital of Beijing PLA were enrolled, and 26 healthy volunteers were served as controls. The expression of γ-GCS in colonic mucosa was determined by immunohistochemistiy. Results: γ-GCS was mainly expressed in the glandular cell cytoplasma. The expression of γ-GCS in UC patients was significantly lower than that in controls (53.3% vs. 92.3%, P〈0.01). Conclusions: The expression of γ-GCS in UC patients is significantly decreased, which suggests that the disequilibrium of oxidative stress exists in colonic colonic muecosa of UC patients, and the low expression of γ-GCS is relevant with the decrease of synthesis of GSH.
