猪血红蛋白寡肽对内皮素Ⅰ的分泌抑制作用研究

摘　　要：目的探讨珠蛋白寡肽对猪主动脉血管内皮细胞(PAEC)的内皮素-1(ET-1)分泌水平的影响。方法本研究采用ELISA方法。结果猪主动脉内皮细胞在正常培养下,内皮素的含量是18.1μg/ml。珠蛋白寡肽组分Ⅱ、Ⅲ、Ⅳ、Ⅴ作用48h后,内皮素含量分别减少了13.9%,13.1%,15.2%,17.7%。结论研究表明珠蛋白胃蛋白水解产物,经SephadexLH-20柱层析分离后得到的5个寡肽组分中,组分Ⅱ、Ⅲ、Ⅳ、Ⅴ对PAEC的ET-1的分泌有抑制作用。Objective To determine the effect of globin oligopeptide on porcine aortic endothelial cells （PAEC） of endothelin-1 （ET-I） secretion levels. Methods ELISA method was used. Results The endothelin content was 18.1 μg/ml while the porcine aortic endothelial cells under normal culture. After the globin oligopeptide components Ⅱ , Ⅲ, Ⅳ, Ⅴwas cultured for 48 h, the level of ET decreased by 13.9%, 13.1%, 15.2%, 17.7%. Conclusion Among 5 oligopeptide components that obtained after peptone hydrolyzates of globin protein chromatographic separated by SephadexLH-20 column, the components Ⅱ , Ⅲ, Ⅳ, Ⅴ can inhibit ET-lsecretion of the PAEC.
