杜仲糖类成分的气相色谱-质谱联用分析

摘　　要：目的研究杜仲的糖类成分,为杜仲开发利用提供实验依据。方法提取杜仲的游离糖成分和多糖成分,用三氟乙酸将多糖水解,依次加入盐酸羟胺、吡啶和乙酸酐进行乙酰化反应,测定杜仲游离糖成分和多糖成分的GC-MS图,分析杜仲的糖类成分。结果通过质谱分析,确定杜仲含有D-木糖、D-核糖、D-阿拉伯糖、D-木糖醇、D-呋喃葡萄糖,α-D-吡喃葡萄糖,β-D-吡喃甘露糖、葡萄糖醇、肌醇、纤维二糖、曲二糖和蔗糖等糖类成分。结论杜仲中的糖类成分种类丰富,气相色谱和质谱联用技术能够有效地分析杜仲的糖类成分。Objective To study the carbohydrate components of Cortex Eucommiae and provide experimental evidence for the exploitation of Cortex Eucommiae.Methods The free sugar and polysaccharides of Cortex Eucommiae were extracted,then the polysaccharides were hydrolyzed with trifluoroacetic acid（TFA） and followed by adding hydroxylamine hydrochloride,pyridine and acetic anhydride acetylation for reaction.After that,Gas chromatography-mass（GC-MS） spectrometry of free sugar and polysaccharides was seasured.Results The carbohydrate components of Cortex Eucommiae consist of D-xylose,D-ribose,D-arabinose,D-xylitol,D-glucofuranose,α-D-glucopyranose,β-D-mannopyranose,D-glucitol,cellobiose,kojibiose and sucrose.Conclusion There are many kinds of carbohydrates in Cortex Eucommiae and the GC-MS method can analyze the sugar composition of Cortex Eucommiae effectively.
