摘  要:目的采用双向聚丙烯酰胺凝胶电泳技术,建立椎动脉缺血型颈椎病肝阳化风证和肝阳上亢证高分辨率的二维凝胶电泳图谱,从蛋白质组学角度探讨椎动脉缺血型颈椎病中医肝阳化风证和肝阳上亢证的本质内涵。方法设颈椎病肝阳化风证组、颈椎病肝阳上亢证组及健康人对照组3组,取外周静脉血分离淋巴细胞提取蛋白质,经双向凝胶电泳,考马斯亮蓝染色获取凝胶图谱,以PDQuestV7.3.1软件对三组表达图谱进行差异分析,选定差异蛋白点进行质谱分析和SWISS-PROT数据库检索鉴定蛋白质。结果建立了颈椎病肝阳化风证组、肝阳上亢证组、健康人组外周静脉血淋巴细胞2-DE图谱;通过分析比较,发现16个蛋白质点在肝阳化风证组中异常表达,12个蛋白质点在肝阳上亢证组中异常表达,肝阳化风证组与肝阳上亢证组比较,分析发现16个差异蛋白点,3个相同蛋白质点。结论颈椎病(椎动脉缺血型)肝阳化风证与肝阳上亢证有差异蛋白又有相同的蛋白,提示同病异证在共同的物质基础上存在着不同的本质内涵。Objective With the technology of two- dimensional polyacrylamide gel electrophoresis (2 - DE), to establish high resolution 2 - dimensional electrophoregram of Gan Yang Hua Feng syndrome (GYHF) and Gan Yang Shang Kang syndrome (GYSK) of cervical spondylosis of Vertebral artery, using proteomics to explore the essence of GYHF and GYSK syndrome of cervical spondylosis of vertebral artery. Methods Three groups were designed, including GYHF of cervical spondylosis of vertebral artery, GYSK of cervical spondylosis of vertebral artery, and healthy control group. The total proteins were extracted from lymphocytes of peripheral venous blood, and two- dimensional electrophoresis was used to separate total proteins. Matrix- Assisted Laser Desorption/Ionization- Time of Flight Mass Spectrometry (MALDI - TOF - MS) and searching in SWISS- PORT and MSDB database were used to identify proteins. Results We obtained high resolution 2 - dimensional electrophoregrams of GYHF, GYSK and healthy people groups. After conducting analysis, 16 differential expressing proteins were found in the GYHF electrophoregram and 12 in GYSK electrophoregram. 16 different protein spots and 3 identical protein spots were observed between GYHF and GYSK groups. Cenclusiens The GYHF and GYSK of cervical spondylosis of vertebral artery have different proteins and identical proteins, which indicate that the two syndromes of the same disease have the same matter substructure and different essential connotation.
