摘  要:[目的]探讨正常大鼠腰椎骨组织成骨细胞和破骨细胞的体外培养方法。[方法]分离成年大鼠腰椎(L1~5),分别采用酶消化法分离获取成骨细胞,骨髓诱导法分离获取破骨细胞,并对在体外培养的成骨细胞和破骨细胞进行鉴定与功能评价。[结果]所培养的成骨细胞表达标志性分化基因,具有较好的钙化能力;破骨细胞表现为多核巨细胞外观,抗酒石酸酸性磷酸酶染色阳性,具有良好的骨吸收功能。[结论]成功分离、培养获得了成年大鼠腰椎成骨细胞和破骨细胞,为进一步研究骨质疏松腰椎并发症的预防与治疗方法打下基础。[ Objective ]To establish the Culture methods for osteoblasts and osteoclasts of the lumbar vertebra of normal adult rats. [ Methods] Lumbar vertebra of adult female SD rats were Chosen, then osteoblasts and osteoclasts were harvested through the enzyme digestion method and bone-marrow'cells induced differentiation approach, respectively. The in vitro cul' tured osteoblasts were detected by RT-PCR assay, and the mineralized function were also observed. Furthermore, the in vitro cultured osteoclasts were identified by morphological observation, toluidlne blue staining, Faefin ring observation, tartrate-resistant acid phosphatase (TRAP) staining, bone resorption assay, etc. E Results~ The in vitro euhured osteoblasts expressed osteoblasfic marked genes and were available with good mineralized ability. The in vitro cultured osteoelasts were found appearance of multinueleate cells with bright-region structure and positive stained TRAP, with ability of bone resorptive pit formation. [Conculsion] The in vitro cultured osteoblast and osteocalst of lumbar vertebra Of adult normal SD rats can be available for following experiments for earlierly preventing and curing relative lumbar vertebra complications of osteoporosis.
