摘  要:目的观察全脑缺血/再灌注损伤大鼠海马组织中PPARαmRNA和蛋白表达的动态变化。方法采用夹闭两侧颈总动脉,颈静脉抽血再回输建立大鼠全脑缺血/再灌注模型(I/R)。RT-PCR和Western Blot分别检测PPARαmRNA和蛋白在缺血再灌注不同时间的表达。结果大鼠海马PPARαmRNA表达在缺血/再灌注30min后升高,24h时达峰,而后降低,再灌注30d仍略高于正常水平。PPARα蛋白表达变化与PPARαmRNA表达相似。结论全脑缺血/再灌注损伤可诱导PPARα mRNA及蛋白表达,升高时限为30d。Objective To explore the changes of PPARα mRNA and protein expression in cerebral hippocampi of global cerebral ischemia reperfusion(I/R) rats. Methods The model of global cerebral ischemia reperfusion was established by bilateral common carotid arteries occlusion and vena cervicalis blood retransfusion. RT-PCR was performed to detect expression of PPARα mRNA. Western Blot was used to detect PPARα protein expression. Results The expression of PPARα mRNA in hippocampi increased at 30 min after global cerebral/JR, peaking at 24 h, and was still slightly higher than normal levels on the 30th day after I/R. The changes of PPARα protein expression were similar with that of PPARα mRNA expression. Conclusion Global cerebral I/R can induce an increase of expression of PPARα mRNA and protein in rat hippocampi within 30 days after insult.
