兔VX_2肿瘤淋巴转移动物模型建立和病理学观察

摘　　要：目的建立适用的VX2淋巴癌转移模型。方法 24只新西兰白兔随机分成模型组和对照组,模型组14只,对照组10只。模型组在兔右股二头肌肌腹中部注射VX_2细胞悬液0.5mL,对照组在相同位置注射0.5mL生理氯化钠溶液,观察淋巴结的变化,第28天处死动物,病理学观察肿瘤大小、形态及组织学改变。结果模型组14只动物全部成瘤,成瘤率100%。腘窝淋巴结明显肿大,病理检查显示其淋巴结可见大量癌细胞,淋巴结的正常结构完全被破坏,甚至消失,转移率100%。结论肌肉注射VX2组织悬液能建立腘窝淋巴结癌转移动物模型,方法简便易行,转移率高。该模型动物体形较大,淋巴结浅而大,对实验性癌淋巴转移的研究具有实用意义。Objective To establish the applicable ttansfer model of VX2 carcinoma lymph node metastases. Methods 24 New Zealand white rabbits were randomly divided into a model group and control group （ model group = 14, control group = 10）. In the model group, the right biceps femoris muscle in the middle of Department was injected with a 0.5 mL VX2 cell suspension. The same location in the control group was injected with 0.5 mL 0.9% sodium chloride solution. The changes in lymph nodes were observed the first 28 days animals were killed. Tumor size, shape and histological changes were observed. Results All model group animals had tumors, and the tumor formation rate was 100%. Popliteal lymph nodes enlarged obviously and pathological examination showed that the lymph node had a large number of cancer ceils; the normal structure of the lymph nodes was completely destroyed, or even disappeared, and the transfer rate was 100%. Conclusion I. M VX2 tissue suspension can establish metastasis in an animal model of popliteal lymph node. The method is simple and easy to use and the transfer rate is high. The model animal shape is larger and the lymph nodes are shallow and large. This model is significant for research in lymph node metastasis in experimental cancer.
