摘 要:目的研究高铁饮食对C57BL/6小鼠黑质(SN)和中脑腹侧被盖区(VTA)多巴胺能神经元的不同损伤。方法将C57BL/6小鼠随机分为高铁饮食组和正常对照组,利用免疫组织化学染色法及铁染色技术检测两组小鼠SN和VTA的酪氨酸羟化酶(TH)阳性细胞数及铁水平的变化。结果高铁饮食组SN的TH阳性细胞数较正常对照组明显减少(t=16.31,P<0.001),铁染色阳性细胞数较对照组明显增高(t=13.01,P<0.001)。而两组小鼠VTA的TH阳性细胞数及铁染色阳性细胞数均无明显差别。结论铁在SN的选择性聚积参与了SN多巴胺能神经元的选择性损伤。Objective To investigate the effect of ferric diet on the degeneration of dopaminergic neurons between the substantia nigra(SN) and the ventral tegmental area(VTA) in C57BL/6 mice. Methods The male C57BL/6 mice were randomly divided into two groups: ferric-diet group and normal-diet group.Tyrosine hydroxylase(TH) positive cells and iron positive cells in SN and VTA were determined using immunohistochemistry and iron staining. Results Compared with those of the control group,the number of TH-positive cells in SN was significantly decreased(t=16.31,P0.001) and the number of iron-positive cells significantly increased(t=13.01,P0.001) in ferric-diet group.The numbers of both TH-positive cells and iron-positive cells were not significantly different in the VTA between the two groups. Conclusion Selective iron accumulation in SN is involved in the selective degeneration of dopamine neurons in SN.
