宫颈癌组织calpain 6表达及其与微血管密度关系

摘　　要：目的研究钙激活中性蛋白酶6(calpain 6)在宫颈癌组织中的表达及其与微血管密度(MVD)的相关性。方法应用免疫组化PV-9000通用二步法,检测calpain 6在10例正常宫颈组织、30例宫颈上皮内瘤变(CIN)及65例宫颈癌组织中的表达并计数MVD。结果 calpain 6在正常宫颈、CIN、宫颈癌组织中的阳性表达率逐渐升高(H=30.05,P<0.01);宫颈癌组织中,临床FIGO分期晚、分化程度差、有淋巴结转移者calpain 6表达明显高于临床分期早、分化程度好和无淋巴结转移者(χ2=4.068～9.524,P<0.05),其表达与病人年龄、组织学类型无关;calpain 6表达与MVD呈显著正相关(r=0.946,P<0.01)。结论 calpain 6可能通过增加肿瘤新生血管形成促进宫颈癌的生长、浸润和转移。Objective To investigate the expression of calpain 6 in cervical intraepithelial neoplasia（CIN） and cancer of the cervix（CC） and analyze its association with microvessel density（MVD）. Methods Calpain 6 expression was observed byusing immunohistochemistry PV-9000 technique and MVD counted in 10 cases of normal cervical epithelium（NCE）,30 of CIN,and 65 of CC. Results The positive rates of calpain 6 expression in NCE,CIN and CC gradually increased（H=30.05,P0.01）.The expression was significantly higher in patients with cancer at late FIGO stage,with poor differentiation and lymph node metastasis than that in patients at early stage,with good differentiation and without lymph node metastasis（χ2=4.068-9.524,P0.05）.The expression of calpain 6 had no significant association with patient＇s age and histology type,but was positively correlated with MVD（r=0.946,P0.01）. Conclusion The results suggested that calpain 6 enhances tumor growth,infiltration and metastasis via an increase of tumor neovascularity.
