缬沙坦对糖尿病大鼠视网膜组织NF-κB表达影响

摘　　要：目的探讨缬沙坦对糖尿病大鼠视网膜组织核因子-κB（NF-κB）表达的影响。方法将20只Wistar大鼠随机分为正常对照组、高糖诱导视网膜病变模型组、缬沙坦处理组和缓冲液处理组,每组5只大鼠。正常对照组和模型组不做处理,缬沙坦组给予缬沙坦40 mg/kg灌胃,缓冲液组给予相同体积缓冲液灌胃,用免疫组织化学法检测视网膜组织中NF-κB的表达水平。结果正常对照组大鼠视网膜组织NF-κB无阳性表达,模型组、缬沙坦组、缓冲液组NF-κB表达水平均明显高于正常对照组（F=262.00,q=18.06~34.59,P〈0.01）;缬沙坦组NFκ-B表达水平较模型组明显降低（q=15.29,P〈0.01）。结论缬沙坦能降低大鼠视网膜组织中NF-κB表达水平,对糖尿病大鼠视网膜病变具有保护作用。Objective To observe the effect of Valsartan on NF-κB expression in retina of diabetic rats. MethodsTwenty Wistar rats were equally randomized to four groups：control group,high-glucose-induced retinopathy（HGIR） group,buffer group and Valsartan group.The control group and HGIR group received no therapy.The mice in Valsartan group were given Valsartan（40 mg/kg） intragastrically.Those in buffer group were given the same volume of buffer as Valsartan group intragastrically.The expression of NF-κB in retinal tissue was examined immunohistochemically. Results There was no expression of NF-κB in normal retina tissue.The expression of NF-κB in the other three groups was significantly higher than the control group（F=262.00;q=18.06-34.59;P0.01）,while the NF-κB expression in Valsartan group was significantly lower than that of the HGIR group（q=15.29,P0.01）. Conclusion Valsartan provided potential protection against retinopathy in diabetic rats via reducing NF-κB expression of retinal tissue.
