摘 要:为了解棉花腺体形成时期相关基因的表达谱,分别抽提陆地棉湘棉-18和N5在腺体形成时期的RNA,采用Affymetrix芯片,扫描采集数据后进行分析。结果显示,在腺体形成时期有明显差异表达的基因共有514条,包括209条上调基因和305条下调基因;在514条差异表达基因中有275条(53.5%)与GenBank中已知的基因具有较高的相似性,这些基因主要涉及防御反应及异戊二烯生物合成等,有239条(46.5%)在GenBank中无任何注释。KOBAS分析结果显示,这些差异基因在KEGG数据库上涉及19条生化途径。说明,腺体形成过程是一个复杂的基因网络调控过程,涉及多条萜类化合物生物合成途径及其他与防御基因相关的途径。To identify gene expression patterns of pigment gland morphogenesis in cotton,genes involved in pigment gland morphogenesis in upland cotton varieties Xiang-18 and N5 were profiled using Affymetrix cotton gene chip.The results showed that there were 514 genes expressed differentially during gland morphogenesis,of which 209 genes were up-regulated and 305 genes were down-regulated.Out of the 514 genes,275(53.5%) genes shared high similarity in nucleic sequence with known genes in GenBank,and the remained 239(46.5%) genes were unknown.The known genes were mainly involved in defense response and isoprenoid biosynthetic process.The KOBAS(KEGG Orthology-Based Annotation System) analysis indicated that the differentially expressed genes were involved in 19 metabolic pathways.The results suggested that pigment gland morphogenesis in cotton was regulated by a complicate gene network,including several terpenoid biosynthetic pathways and plant defense-related pathways.
