摘  要:目的探讨cytokine-TLR-SOCS通路在原发性胆汁性肝硬化(primary biliary cirrhosis,PBC)发病机制中的作用。方法研究对象分为3组:32例PBC,30例慢性乙肝(chronic hepatitis B,CHB)和32例健康人(healthy controls,HC)。ELISA测定血清IL-6和IFNγ的变化,RT-PCR测定外周血单个核细胞的TLR和SOCS的表达。结果与HC组相比,PBC组的IL-6和IFNγ水平显著升高;PBC组的TLR4、TLR9、SOCS1和SOCS3 mRNA的水平均显著性高于HC,为HC组的1.48、2.51、3.43和1.48倍,TLR9和SOCS1的mRNA水平是CHB组的6.85倍和2.99倍(P<0.05)。结论 PBC患者处于炎性状态;TLR的过度激活可能与PBC的发生和发展有一定关系,SOCS作为通路的负调控蛋白可作为疾病免疫治疗的靶点。Objective To study the role of cytokine - TLR - SOCS in the development/progression of primary biliary cirrhosis (PBC). Methods Thirty-two patients with PBC, 30 patients with chronic hepatitis B (CHB), and 32 healthy controls (HC) were en- rolled in this study. The serum concentrations of IL - 6 and IFN3, were determined by enzyme - linked immunosorbent assay. In addition, THE expression of TLR4, TLR9, SOCS1 and SOCS3 genes in the peripheral blood mononuclear cells of patients were assayed by RT - PCR. Results The levels of IL - 6 and IFN7 were significantly elevated in PBC patients compared with HC, the mean TLR4, TLR9, SOCS1 and SOCS3 mRNA levels were significantly higher in the PBC group than those in the HC ( mean 1.48, 2.51, 3.43 and 1.48 folds, P 〈 0.05 ). THE expressions of TLR9 and SOCS1 in PBC group were 6.85 and 2.99 times those in CHB group, respectively. Conclusions Inflammatory status exists in the progression of PBC. TLR plays an important role in the pathogenesis of PBC, SOCS as an important negative regulator of TLR signaling, maybe a potential target of new immunotherapy for PBC.
