胃癌中管家基因表达的恒定性研究

摘　　要：目的观察胃癌中管家基因的表达是否恒定。方法通过分析胃癌基因表达序列分析数据库(SAGE),观察胃癌中管家基因的表达是否恒定。同时运用逆转录多聚酶链反应(RT-PCR)分别测定胃癌组织、正常组织中管家基因GAPDH、β-actin mRNA的表达情况,并进行半定量灰度分析,对结果进行验证。结果通过分析胃癌SAGE数据库中管家基因的拷贝数,发现在不同病变组织以及病变组织与正常组织间多数管家基因的表达是不恒定的。GAPDH、β-ac-tin在胃癌中的表达发生明显改变,与正常组织比较,差别均有统计学意义(P<0.01)。结论胃癌中管家基因表达的不恒定性普遍存在。由于管家基因在胃癌中的表达不恒定,在运用RT-PCR技术对胃癌基因表达进行定量分析时,应选择适当的2种或2种以上的内参基因,以减少检测标本间的差异。Objective To investigate the stability of housekeeping genes expression in gastric cancer.Methods The database of serial analysis of gene expression（SAGE）was used to investigate the stability of housekeeping genes expression in gastric cancer.Meanwhile,the reverse transcription-polymerase chain reaction（RT-PCR）was taken to measure the expression of housekeeping gene GAPDH and β-actin mRNA in gastric cancer tissue and normal control tissue,and the electrophoretic bands were analyzed by semiquantitative.Results The copy numbers of the housekeeping genes in gastric cancer SAGE database indicate that the expression levels of the same housekeeping genes in different cancerous tissues,and also between the cancerous and normal tissues is variable.The expression levels of GAPDH and β-actin in the gastric carcinoma tissues were variable,and compared with nomal tissue,there were statistic difference（P0.01）.Conclusion Housekeeping gene expressions are not always constant in gastric carcinoma.It should be careful to select an endogenous reference when RT-PCR assays is used to analyse gastric carcinoma gene expressions.As housekeeping gene expression in gastric cancer is not constant,two or more than two endogenous reference should be selected to reduce the differences between test specimens in quantitative experiments.
