葛根素对局灶脑缺血大鼠脑组织内LPO含量影响的研究

摘　　要：目的:探讨大鼠局灶性脑缺血不同脑组织中脂质过氧化物(LPO)的表达及葛根素的保护作用。方法:采用线栓法制备大鼠大脑中动脉持续局灶性脑缺血模型,在缺血后2h、12h、24h给予葛根素进行干预,运用TTC染色观察脑梗死体积的变化,荧光探针乙酰乙酸盐-2,7-二氯氢化荧光素(2,7-dichlorofluorescindiacetate,D-399)标记,激光共聚焦扫描显微镜观察不同组别活体脑片细胞内LPO含量的变化。结果:2h和12h干预组脑梗死体积明显小于对照组,24h干预组与对照组比较,梗死体积无统计学差异;缺血2h组梗死侧皮质、纹状体区LPO含量增加,随缺血时间的延长,荧光强度逐渐增强,24h组最强,与假手术组比较差异显著;干预组与对照组比较皮质和纹状体区荧光强度均下降,有统计学意义。结论:脑缺血后早期即出现脂质过氧化损伤,随时间延长损伤逐渐加重,应用葛根素可通过减轻脂质过氧化损伤起到脑保护作用。Obiective：To investigate the expression of lipid peroxidation （LPO） on continued focal cerebral ischemia and the protection of puerarin flavonoids in the rats.Methods：Continued focal cerebral ischemia model was set up with middle cerebral artery occlusion by intraluminal block in this study.After cerebral ischemia,puerarin was administered at 2h、12h、24h.The volume of cerebral ischemia was assessed by TTC dyeing.The fluorescence intensity of LPO in living brain slices cell of different groups,marked by fluorescent probe of 2,7-dichlorofluorescin diacetate（D-399）,was determined under laser scanning confocal microscope.Results：The volumes of cerebral ischemia were decreased significantly in 2h and 12h puerarin treatment groups,compared with the control.There was no significant difference in the 24h groups.The fluorescence intensity of LDP in infarction cortex and corpora striata begun to step up significantly in 2h group after ischemia and further enhanced with the prolonged ischemic time in ischemia model rats.Compared with the control,the intensity decreased significantly at the two regions in the all three puerarin treatment groups in different time.Conclusion：Lipid peroxidation increase was seen in the early stage of cerebral ischemia,and enhanced further with the prolonged ischemic time.The treatment of puerarin can relieve LPO overload and reduce the volume of cerebral ischemia and play a neuroprotective role in the early time.
