摘  要:目的:观察大鼠磨牙氢氧化钙直接盖髓术后诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)的表达变化。方法:42只雌性wistar大鼠第一磨牙机械穿髓后用氢氧化钙直接盖髓,分别于术后即刻、1、3、7、14、21、28d处死动物并取材,进行免疫组织化学染色,观察iNOS在牙髓组织的动态表达和定位情况,用图像分析软件测定各组标本中iNOS阳性染色的平均光密度值(optical density,OD)。结果:在正常牙髓组织中iNOS表达呈阴性,氢氧化钙直接盖髓后不同时间iNOS表达呈弱阳性-阳性-弱阳性-阴性的变化,其中盖髓后3diNOS表达最强,21d接近正常水平。表达主要位于盖髓剂下方中性粒细胞、成纤维细胞、成牙本质细胞、血管内皮细胞等。结论:氢氧化钙直接盖髓后,iNOS表达随时间变化呈现先增高后降低的趋势,表达强度与牙髓组织修复程度有关。AIM : To observe the expression of inducible nitric oxide synthase (iNOS) in rat dental pulp after direct pulp capping with calcium hydroxide. METHODS: Forty-two female Wistar rats were included in this study. The first molars on one side of the maxilla and mandible were mechanically exposed and the exposed dental pulps were directly capped with calcium hydroxide. The contralateral molars received no treatment and served as con- trols. The rats were sacrificed immediately, 1 d, 3 d, 7 d,14 d, 21 d and 28 d after direct pulp capping, iNOS ex- pression in pulp tissue was detected with itnmunohistochemical staining. RESULTS: No iNOS expression was detected in the control group. After direct pulp capping with calcium hydroxide, iNOS expression gradually increased and reached peak expression on 3 d and 5 d post-direct capping ( P 〈 0.05 ). iNOS expression gradually decreased after- wards and reached normal level on 21 d post-direct capping, iNOS expression was mainly detected in fibroblasts, odon- toblasts cells, vascular endothelial cells and neutrophils. CONCLUSION: After direct pulp capping with calcium hy- droxide, a time-dependent iNOS expression change was observed. Our results suggest that iNOS expression is related with the process of dental pulp repair.
