摘 要:目的探讨松果体、中枢核团视交叉上核(SCN)和中缝背核(DR)与外周淋巴细胞之间昼夜节律的联系和传递关系。方法在标准的光照条件下,测定中枢和外周第一信使褪黑素(MT)、第二信使cAMP和cGMP的昼夜节律。结果在小鼠松果体和血清中,MT含量的昼夜节律峰值位于黑暗中期,而各组织中cAMP/cGMP比值的节律峰值却位于光照中期,呈现相互倒置的位相关系。各组织中cAMP/cGMP比值的昼夜节律的峰值分别为:松果体,-155.82°(CT10 :23);SCN,-166.30°(CT10:54);DR,-179.16°(CT11:56);血清,-186.66°(CT12:27);淋巴细胞,-188.00°(CT12:32)。结论依据cAMP/cGMP峰值出现的昼夜时间先后,由中枢向外周淋巴细胞的信息传递按照松果体→SCN→DR→血液→淋巴细胞的顺序进行。To explore the transductive relationship between pineal, central nuclei of suprachiasmatic nucleus (SCN) and dorsal raphe(DR), and peripheral lymphocyte in terms of circadian rhythm. Methods Circadian changes of melatonin (MT), cAMP and cGMP contents in the central nuclei and lymphocytes of mice were determined under the standard light-dark cycle. Results The peak of MT rhythm in pineal and serum was found at middle of the dark phase, and that of the cAMP/cGMP ratio in each tissue examed was presented at middle of the light phase, that is: pineal, - 155. 82°(CT10 : 23) ; SCN, - 166. 30°(CT10:54);DR, - 179. 16°(CT11 : 56); serum, - 186. 66°(CT12 : 27) ;and lymphocyte, - 188. 00°(CT12 : 32),respectively. The peaks of MT and the cAMP/cGMP ratio showed an inverse phase relationship. Conclusion The sequence of the signal transduction betweem the central nuclei and the peripheral lymphocytes could be traced from pineal to SCN, then to DR and serum, and finally to lymphocytes, according to the timepoints at which the peak phase of the cAMP/cGMP rhythms appeared in a circadian day.
