精氨酸对延边黄牛体细胞克隆重组胚发育的影响

摘  要:为了提高延边黄牛体细胞克隆重组胚的发育率,试验研究了在体外培养液中添加不同浓度的精氨酸对重组胚发育的影响。选择形态良好排出第一极体的卵母细胞进行去核与注核操作,然后开始融合、激活,最后在添加不同浓度(0、0.061、0.132、0.264mmol/L)精氨酸的培养液中培养。结果发现,对照组与处理组之间重组胚的卵裂率和8-16C的发育率差异不显著,而囊胚的发育上,0.061mmol/L处理组显著高于对照组。表明体外培养液中添加精氨酸后能够促进体细胞克隆重组胚的发育。In order to improve the development rate of somatic cell cloned reorganization embryo, the effect of different Arginine concentration added to in vitro culture medium on the development of reorganization embryo was studied. The first polar body from oocytes was used to enucleate, inject nuclear, amalgamate, activate, and finally cultivate in culture medium with different concentrations (0 mmol/L,0.061 mmol/L,0.132 mmol/L,0.264 mmol/L) of Arginine. Results showed that between the control group and experimental group, there was no significant difference in cleavage rate of recombinant embryoes and the development rate of 8-16C, and the development rate of blastocyst at the concentration of 0.061 mmol/L Arginine was significantly higher than that of the control group. So adding Arginine in vitro culture medium could promote the development of cloned recombinant embryoes.
