摘  要:目的:分析低氧条件对肝癌细胞化疗敏感性的变化,从细胞水平研究低氧环境对化疗药物诱导的肝癌细胞凋亡的影响。方法:测定不同浓度化疗药物(CDDP、EPI、GEM、MMC)对细胞死亡率的影响及IC50值,对人肝癌细胞SMMC-7721化疗药物处理,经过常氧及低氧不同环境培养下,分析肝癌细胞死亡率以及凋亡情况。结果:实验结果显示,化疗药物作用浓度与细胞死亡率正相关;低氧培养条件下化疗药物引起的细胞死亡率明显低于常氧培养组,两组有显著差异(P<0.05);低氧环境下化疗药物诱导的SMMC-7721细胞凋亡率明显降低(P<0.05)。结论:低氧环境下肝癌细胞对化疗药物的不敏感与低氧引起肝癌细胞凋亡能力下调有关。Objective:To analyze the changes of sensitivity of hepatocellular carcinoma cells to chemotherapeutic dugs in hypoxia.Meanwhile,to find the effects of hypoxia on chemotherapeutic drug-induced apoptosis of hepatocellular carcinoma cells by the cellular level.Methods: The different concentration of chemotherapeutic drugs(cisplatin,epirubicin,gemcitabine and mitomycin) effect on hepatocellular carcinoma cell SMMC-7721,then got the cell death ratios by MTT method,also calculated their IC50 value.SMMC-7721 in both environment of 20% oxygen and 1% oxygen adding those chemotherapeutic drugs,and analyzed the rate of cell death and apoptosis.Results: There was a correlation between the concentration of chemotherapeutic drugs and cell death ratios.Compared with normal oxygen group,cell death ratios were lower in hypoxic group(P0.05).As well as the rates of apoptosis were lower after chemotherapeutic drug-induced in hypoxia environment(P0.05),there were significant statistical differences.Conclusions: In hypoxic environment,the insensitive to chemotherapeutic drugs of Hepatocellular carcinoma cells is due to the down-abilities of apoptosis.
