摘 要:Dmrt(doublesex and mab-3 relatated transcription factors,Dmrt)基因家族是在动物中发现的一类编码转录因子,该家族成员的共同特征是含有1个高度保守的DM结构域,其能识别特异DNA序列,在性别决定和分化发育的调控中担负重要的功能。本研究参照不同物种DM结构域设计了1对简并引物,扩增和克隆了尼氏真绥螨[Euseius nicholsi(Ehara et Lee)]Dmrt基因的DM结构域,获得了4个不同的DM结构域克隆,分别命名为EnDm-rt2a、EnDmrt2b、EnDmrt2c、EnDmrt2d。与不同进化地位的其他动物Dmrt基因的DM序列进行比对和聚类分析,结果显示DM序列及其编码氨基酸具有高度的相似性,表明DM及Dmrt基因在系统进化上具有高度的序列保守性和功能保守性。The Dmrt gene family, doublesex and mab -3 relatated transcription factors, are found throughout the animal kingdom. They are encoding transcription factors and characterized by a conserved zinc fingerlike DNA - binding motif, the DM domain. They play an impotent role in the sexual determination in development. In this paper, degenemte PCR was used to amplify the conserved DM domains of Dmrt genes in Euseius nicholsi. Four different DM sequences were identified and named as EnDmrt2a,EnDmrt2b,EnDmrt2c, EnDmrt2d separately. The comparative and phylogenie analysis revealed that the amino acid sequences encoded by DM of Dmrt genes from different species are highly homology. The results indicated that the DM and the Dmrt genes were highly consenrative in sequence and function in evolutionary process.
