榄香烯乳对人肺腺癌A549细胞株细胞周期及凋亡的影响

摘　　要：目的:探讨榄香烯乳对人肺腺癌A549细胞株细胞周期及凋亡的作用。方法:体外培养人肺腺癌A549细胞株,四甲基偶氮唑蓝比色法测定不同浓度榄香烯乳在不同作用时间下对A549细胞株的生长抑制作用。用流式细胞术检测榄香烯乳作用24h后A549细胞株的周期及凋亡变化。结果:榄香烯乳作用后人肺腺癌A549细胞株增殖明显受抑制,呈时间和剂量依赖性。作用24h后G2/M期比例明显升高,S期细胞比例显著下降,凋亡率增加。结论:榄香烯乳对人肺腺癌A549细胞株有明显的增殖抑制作用,可引起A549细胞G2/M期阻滞及诱导其凋亡。Objective：To explore the influence on the cell cycle and apoptosis of human lung adenocarcinoma A549 cell line managed by Elemene. Methods：Human lung adenocarcinoma A549 cell line was cultured in vitro. The growth inhibiting effects of different concentrations of Elemene on A549 cells at different times were measured by MTT colorimetric test. The cell cycle and apoptosis changes of A549 line were observed by flow cytometry analysis after Elemene managed 24h. Results：The proliferation of human lung adenocarcinoma A549 cell line was inhibited significantly by Elemene and the inhibition was time and dose-dependent. After Elemene managed 24h, G2/M phase fraction was high significantly, while the Sphase fraction was decreased significantly and apoptosis rate increased. Conclusions：Human lung adenocarcinoma A549 cell line was inhibited significantly by Elemene the cell cycle of A549 was blocked in G2/M phase and apoptosis was induced by Elemene.
