摘 要:为了确定从酸枣仁中提取、纯化皂苷A、B的最适方法和最佳工艺条件,以正交试验优化并比较回流法和超声法提取皂苷的工艺条件,筛选一种适合纯化酸枣仁皂苷A、B的树脂,确定上柱试验的最佳工艺参数。试验结果表明:超声法较回流法的提取效果好,其最佳提取条件是:50%(体积分数)乙醇溶液,液固比(mL:g)25:1,功率750W,50℃提取0.5h。SP700树脂具有最大的吸附容量和解吸得率,将其装柱后做上柱试验,用50%乙醇和90%乙醇进行梯度洗脱,皂苷A、B被完全洗脱出来,小柱和大柱的结果具有很好的重复性。To obtain the best appropriate method and optimum conditions of extraction and purification of jujuboside A and B from Zizyphi spinosi semen(ZSS),the conditions of reflux and ultrasonic extraction were optimized by orthogonal design respectively.Mitsubishi SP700 resin was selected from 7 different types of macroporous adsorption resins and the elution conditions of chromatography experiment were optimized.Results showed that ultrasonic extraction was better than reflux extraction and the optimum extraction conditions were as follows:50% ethanol solution,ratio of liquid to solid=25:1 (mL/g),ultrasonic power 750 W,0.5 h,50 ℃.SP700 resin was tested with the highest adsorption capacity and desorption rate.In chromatographic experiment using column packed with SP700 resin,jujuboside A and B could be eluted entirely with gradient elution mode of 50% and 90% ethanol solution.And the results between small and scaled up columns were repeated well.
