颈动脉粥样硬化病人血单个核细胞中低密度脂蛋白受体基因表达的研究

摘　　要：研究颈动脉粥样硬化病人外周血单个核细胞低密度脂蛋白受体（ＬＤＬＲ）基因表达与颈动脉粥样硬化程度的相关性。选择颈动脉粥样硬化病人３５例和对照组３１例，采用逆转录ＰＣＲ（ＲＴ－ＰＣＲ）定量测定ＬＤＬＲ的基因表达，以β－ａｃｔｉｎ作为内标物。颈动脉粥样硬化病人外周血单核细胞ＬＤＬＲ基因表达量低于对照组，ＬＤＬＲ基因表达量与血中低密度脂蛋白（ＬＤＬ）水平呈负相关。We have investigated low density lipoprotein receptor(LDLR)gene expression in periphery blood monocytic cells (PBMC)and their relation to carotid atheroslerosis.We studied 35 patient with carotid atherosclerosis and 31 control subjects.We quantified the expression of LDLR gene by reverse transcription polymerase chain reaction (RT-PCR) with β-actin as the internal standard.LDLR gene expression was significantly reduced in the patient group,as compared to the control group.The levels of LDLR gene expression correlated negatively with serum low density lipoprotein (LDL) levels.The reduced expression of LDLR gene is responsible to carotid atherosclerosis.
