摘  要:目的建立尘螨过敏性哮喘小鼠模型,比较不同致敏方式在相同尘螨粗提浸液(CDM)激发下的结果,评价CDM哮喘模型。方法 C57BL/6小鼠随机分成5组,各组分别采用不同的致敏程序致敏,试验第15~18天采用相同剂量与方式的CDM浸液激发。对照组致敏与激发均以磷酸盐缓冲液替代过敏原。从症状、肺泡灌洗液(BALF)的白细胞介素(IL-4)与干扰素-γ(IFN-γ)水平及细胞计数与肺组织病理方面进行评估。结果各模型组从症状,组织病理,BAIF细胞计数、分类(细胞总数增加,其中嗜酸性粒细胞所占比例增大,与对照组比较,P<0.05)及细胞因子检测(IL-4与对照组比较,P<0.05)等均表现出了过敏性哮喘的特点。但是模型组间细胞因子差异无统计学意义。结论各模型组均能够建立小鼠过敏性哮喘模型,证明了CDM的免疫耐受与常规过敏原不同,同时验证了抗原间可能存在协同作用。Objective Built murine asthma model with Crude Dermatophagoides farinae mite extracts(CDM)and compared the index of the different way to sensitized mice after the same CDM challenging,and to assess the CDM model.Methods The C57BL/6 mice were divided into five groups randomly,The mice of 4 experimental groups were sensitized by different methods,From the days of 15 to 18 all the mice except the negative control were given CDM as a challenge.Negative control was treated by PBS.Compare the symptom,the BALF(to test the IL-4,IFN-γ and to count and assort the cell number)and the pathologic changes in lungs.Results Compared to the negative control,all the asthma model groups from the symptom,the organization pathology to the BALF cell counting as well as classified and so on displayed the allergic asthma's characteristic.The BALF total cell counts and eosinophils proportion and IL-4 level compare with the control group,statistically significant differences(P〈0.05).Conclusion This experiment uses CDM,CDM strengthen as well as the OVA to sensitization,then stimulated with the CDM successfully established the mouse allergic asthma model,Which demonstrated that the CDM is different from the other allergen on the immune tolerance,and at the same time demonstrated that between the antigen exists immunity synergism.
